Alvimopan D5 is a deuterated form of Alvimopan, a selective antagonist of the mu-opioid receptor that is used to treat postoperative ileus (POI). POI is a common complication after major abdominal surgery, characterized by delayed gastrointestinal motility that leads to nausea, vomiting, and abdominal distension. Alvimopan D5 works by blocking the effects of opioids on the gastrointestinal tract, thus accelerating the recovery of bowel function after surgery.
Chemically, Alvimopan D5 is a white to off-white crystalline powder that is soluble in water. Its molecular formula is C25H24D5N3O4, and its molecular weight is 436.6 g/mol. Alvimopan D5 is a deuterated form of Alvimopan, which means that some of its hydrogen atoms are replaced by deuterium atoms. This modification does not alter the pharmacological activity of the drug but allows for better detection in pharmacokinetic studies.
The recommended dose of Alvimopan D5 is 12 mg administered orally 30 minutes to 5 hours before surgery, followed by 12 mg twice daily for up to 7 days after surgery. Alvimopan D5 is contraindicated in patients with known or suspected gastrointestinal obstruction or in those who have received therapeutic doses of opioids for more than 7 consecutive days prior to surgery. Common side effects of Alvimopan D5 include abdominal pain, nausea, vomiting, and diarrhea.
