Bivalirudin-(D-Phe-12) is a synthetic peptide consisting of 20 amino acids that is used as an anticoagulant drug. It is a direct thrombin inhibitor that works by binding to the active site of thrombin, thus preventing the formation of fibrin clots. Bivalirudin-(D-Phe-12) is used in patients undergoing percutaneous coronary intervention (PCI) and in those with heparin-induced thrombocytopenia (HIT). It is administered intravenously and has a short half-life of approximately 25 minutes. Bivalirudin-(D-Phe-12) is a modified version of hirudin, a natural anticoagulant found in the saliva of leeches. The modification involves the substitution of phenylalanine at position 12 with D-phenylalanine, which increases the drug's stability and potency. Bivalirudin-(D-Phe-12) also has a lower risk of bleeding compared to other anticoagulants, such as unfractionated heparin. The chemical structure of Bivalirudin-(D-Phe-12) is as follows: H-D-Phe-Pro-Arg-Pro-Gly-Gly-Gly-Gly-Asn-Gly-Asp-Phe-Glu-Glu-Ile-Pro-Glu-Glu-Tyr-Leu-OH. It has a molecular weight of 2180.3 g/mol and a pKa value of 4.5. Bivalirudin-(D-Phe-12) is soluble in water and has a pH range of 5.0-7.5.
